It is a genetic study that evaluates Plasminogen activator inhibitor-1 (PAI-1) The 4G/5G polymorphism in the gene. This polymorphism affects the levels of the PAI-1 protein, which is involved in fibrinolysis and the blood clotting process.
PAI-1 (Plasminogen Activator Inhibitor-1) It is a protein that plays an important role in regulating blood clotting and thrombus dissolution.
The main function of PAI-1 in plasma
PAI-1 is Fibrinolysis inhibitor, that is, it inhibits the process of blood clot dissolution.
Accordingly, elevated levels of PAI-1 in plasma lead to decreased fibrinolytic activity and are associated with the development of arterial or venous thrombosis.
PAI-1 (Plasminogen Activator Inhibitor-1) The protein is synthesized by the PAI-1 gene, which is located on chromosome 7 and contains 8 introns and 9 exons.
The 4G/5G polymorphism of the gene (a guanosine deletion/insertion at position 675 of the PAI-1 gene promoter) is one of the main genetic determinants of PAI-1 levels.
Research material – Venous blood.
One of three possible outcomes is observed for the patient:
In 4G/4G homozygous patients (frequency in the European population approximately 11%) the level of PAI-1 is approximately 25% higher than in 5G/5G homozygotes.
Elevated PAI-1 levels are also found in the heterozygous 4G/5G genotype, the frequency of which in the European population is approximately 40%.
Carrying the 4G allele is associated with obesity, arterial hypertension, and a slightly increased risk of myocardial infarction.
In terms of pregnancy pathology, according to one study, the presence of 4G/4G or 4G/5G genotypes is clinically significant in the development of venous thromboembolism during pregnancy.
The presence of the 4G allele is an independent risk factor for myocardial infarction in young patients (≤45 years), especially in the presence of risk factors (tobacco use, family history of arterial hypertension and cardiovascular diseases, etc.).
In patients with coronary artery disease, the presence of the 4G/4G genotype is associated with an increased risk of sudden death.
The test is not used in isolation. The results are analyzed in conjunction with other clinical and laboratory data.
