Clobazam is a broad-spectrum anticonvulsant used to treat a number of epilepsy syndromes and for migraine prophylaxis.
Clobazam blocks voltage-gated sodium channels, enhances the activity of gamma-aminobutyric acid (GABA) thereby inhibiting glutamate receptors, and the activity of the enzyme carbonic anhydrase – thereby exerting antiepileptic and antimigraine effects.
Clobazam is characterized by good absorption, low protein binding, and minimal hepatic metabolism. It is eliminated from the body by the kidneys, excreted almost unchanged in the urine.
Clobazam has virtually no effect on the plasma concentrations of other anticonvulsants, with the exception of phenytoin, the serum concentration of which increases in the presence of clobazam.
Phenobarbital, phenytoin, and carbamazepine, in turn, reduce the plasma concentration of clobazam.
The most common side effects associated with clobazam include: weight loss, decreased appetite, dizziness, impaired coordination, memory impairment, and paresthesia (sensation changes).
