General Information

The S100 protein family includes 25 proteins with amino acid sequence and structural similarity. However, only a few representatives of these proteins have been studied in depth. S100 proteins are involved in various processes such as: cell proliferation, migration, apoptosis, inflammatory response, Ca2+ homeostasis, energy metabolism and others.
Calcium ions are involved in a number of cellular processes, from muscle contraction to cell differentiation and death. Signaling and intracellular Ca2+ levels are regulated by calcium transporters and membrane channels.
The S100 family of proteins are calcium-binding proteins, although they also bind to zinc and copper. S100 proteins do not have an "intrinsic" function. They perform functions with target proteins, mostly through calcium binding. S100 proteins have both intracellular and extracellular targets.
Due to multiple functions, s100 family proteins may have several names: S100A4 – known as Calvasculin; S100A6 – Calcycline; S100A8/A9 - calprotectin and others.
Some calcium-binding proteins, such as calmodulin and troponin-C, function only at the intracellular level. However, other S100 proteins act both intra- and extracellularly and regulate target cell function in a paracrine and autocrine manner.

S100 proteins as biomarkers of diseases
Since many representatives of S100 proteins are found in various body fluids, they may be used as disease-specific biomarkers.
S100A4 is a biomarker of stem cell glioma, the increased expression of which indicates the presence of metastases.
Elevation of S100A6 protein concentration in serum associated with gastric tumors.
S100A7 is increased in the cerebrospinal fluid and brain of Alzheimer's patients.
Elevation of S100A12 in serum is characteristic of diabetes mellitus. This protein is also a biomarker of inflammation in juvenile idiopathic arthritis.
An increase in S100A8/A9 plasma concentration is characteristic of patients with obesity and coronary artery disease. This protein is also a biomarker for the management of inflammatory bowel diseases, including Crohn's disease. Fecal S100A8/A9 is used to differentiate between inflammatory bowel disease and irritable bowel syndrome.
S100B protein has been established as a biomarker for the assessment of traumatic brain injury. His eyebrow expression is also associated with Down syndrome.

S100 proteins as therapeutic targets
A number of studies have established that S100 proteins can be used as potential therapeutic targets in the treatment of a number of diseases.

S100 proteins - calgranulins play an important role in the immune response of inflammatory diseases (arthritis, atherosclerosis, microbial infections), as well as in joint inflammation and cartilage tissue deformations in patients with rheumatoid arthritis.
S100a7 is abundantly produced in psoriatic lesions.
Several representatives of S100 proteins, mainly S100A4 and S100A8/9, have been identified as key pathogenic components in the development of cancer and diseases such as diabetes and inflammatory diseases.
